Generation of ERE-Kaede-Casper (F0) line. ERE denotes the ERE-Gal4ff transgene sequence, GFP denotes the UAS-GFP transgene sequence and Kaede denotes the UAS-Kaede transgene sequence. Expression of pigmentation (Pig.) genes roy (dark) and nacre (silver) are also shown. The ERE-GFP-Casper model, homozygous for both transgene sequences, and a homozygous UAS-Kaede strain were initially crossed to produce a heterozygous generation. In-breeding within this generation produced progeny with different genotypes based on four genes of interest. At sexual maturity, F0 ERE-Kaede-Casper adults were identified by screening for photoconvertible progeny with fully silenced pigmentation and TG(ERE:Gal4ff)(UAS:Kaede) expression.
