Knockdown of I215L mRNA levels inhibits ASFV DNA replication and progeny production. (A) I215L-depleted cells showed a decreased number of ASFV genomes [1.01 × 109 genomes/ml for siRNA_I sequence (−65.83%) and 1.09 × 109 genomes/ml for siRNA_II (−64.87%)] when compared to the control group (2.98 × 109 genomes/ml, p ≤ 0.05). Results represent the mean of three independent experiments. (B) A statistically significant reduction in viral yield was observed in ASFV-infected Vero cells (MOI of 0.1) transfected with siRNAs against ASFV-I215L (100 nM), in comparison with the GAPDH siRNA-transfected infected cells (between −96.86% and −99.24%; 1 × 104.88 and 1 × 105.50 versus 1 × 107.00 viral particles/ml; p ≤ 0.05), at 72 hpi. The virus yield of each supernatant was calculated from the average of three independent experiments. Error bars represent standard error (SE) of the mean values.
