Amounts of cleaved RNA at various reaction times after treatment with RNase H. 4 μM of the gapmer/RNA duplex (i.e., G^L_smC^L_sa_st_st_sg_sg_st_sa_st_sT^L_smC^L_sA^L/UGAAUACCAAUGC) was treated with 40 U/mL of RNase H in 10 mM Tris buffer containing 100 mM NaCl and 0.5 mM of MgCl₂ at pH 7.2 and 37 °C.
