The NIPS workflow. I-score: two interacting proteins (red and blue) form a complex. The mutation (yellow) is localized with respect to the interface. If the mutation is at the interface, the I-score is 1; otherwise, the score is 0. The T-score is a measure of the proximity of a mutant node to a known cancer node in the protein-protein interaction network compared with the distance to a neutral node. The S-score is derived from the original score predicted by SIFT. The S-score is equal to 1 - SIFT score. A weighted average formula, Condel, was used to integrate these three scores into the ‘meta-score’. All SAPs were ranked according to their meta-scores.
