In vitro stability and pharmacokinetics of HC-scTRAIL. (a) The thermal stability of HC-scTRAIL and Fc-scTRAIL-FAVSGAA was analysed by differential scanning calorimetry. The onset temperatures of unfolding processes are indicated by dotted lines. (b) The bioactivities of HC-scTRAIL and Fc-scTRAIL-FAVSGAA were tested on Colo205 cells after incubating the proteins for different times at 37 °C in 50% human blood plasma (EC₅₀ values normalized to non-incubated control, n = 1, mean of triplicates ± S.D.). (c) The serum concentrations after i.v. administration of 25 µg of HC-scTRAIL in CD-1 mice were analysed by ELISA.
