Real-time polymerase chain reaction (PCR) quantification of fold changes in CCL20, S100A7, S100A8, S100A9, hBD2, and LL-37 transcripts in human primary keratinocytes after the addition of TNFα, IL-17A, and IL-22 recombination proteins. Primary keratinocytes were pretreated with different doses of isoflavone extract for 24 hours and then incubated with TNFα (A), IL-17A (B), and IL-22 (C) for 6 or 8 h. Total RNA was then isolated and mRNA analysis performed via real-time RT-PCR. Data are expressed as fold induction of relevant mRNAs sequences as compared to untreated controls. Data are mean ± SEM from at least three independent experiments performed in triplicate; *P < 0.05 determined by Student’s t-test.
