Western blot analysis of the phosphorylated substrate proteins in signal pathways in hypoxic-BV2 cells after administrated with CGS21680 after 8 h of exposure to low glucose and hypoxia. The upper panel shows the specific band of CREB and p-CREB (43 kD, A), ERK1/2 and p-ERK1/2 (44 kD/42 Kd, B), STAT3 and p-STAT3 (88 Kd, C) and GAPDH (38 kD). The lower panel shows bar graphs showing significant changes in the optical density ratio of phosphor-proteins to that of total proteins between groups after 8 h of exposure to low glucose and hypoxia. The ratio of phosphorylation of CREB in H-89 group and Staurosporine group significantly decreased when compared with VEH group after 8 h of exposure to low glucose and hypoxia. The ratio of phosphorylation of ERK1/2 in Staurosporine group also significantly decreased when compared with VEH group after 8 h of exposure to low glucose and hypoxia. *p < 0.05. The values represent the mean ± SEM in triplicate.
