High-level expression and purification of TNFRII-Trimer fusion protein. (a) 10% SDS-PAGE analysis of TNFRII-Trimer expression from a fed-batch serum-free cell culture in the bioreactor. 10 μl of cell-free conditioned medium from day 7 to day 14 were analyzed under non-reducing condition followed by Coomassie brilliant blue staining. (b) Bioassay analysis of TNFRII-Trimer production in conditioned medium from day 7 to day 14. (c) SDS-PAGE analysis of purified TNFRII-Trimer under either non-reducing or reducing conditions with TNFRII-Fc as a control. TNFRII-Fc (2 μg) and purified TNFRII-Trimer (2 μg) were analyzed by a 10% SDS-PAGE and stained with Coomassie Blue. (d) Purity evaluation of TNFRII-Trimer by SEC-HPLC, with OD280 detection. TNFRII-Fc served as a control. The main peak area of TNFRII-Trimer was 95%.
