Simulating media change protocols to achieve quasi-steady state operations in the gut MPS. Simulating a range of medium change protocols allows identification of operational strategies that can simultaneously satisfy the desired conditions: (a) glucose within physiological levels (between red planes: 4–7 mM) and (b) lactate levels below toxic concentrations (below red plane: 20 mM). Media change protocols simulated were replacement of media volume every 24 h from the basal side (0-100%, 10% steps) and supplementing glucose every 24 h to the apical site (0–10 mM, 1 mM steps) for 10 days, with the final glucose and lactate concentrations represented by the simulated surfaces. The color of the simulated surface corresponds to the colorbar on the right-hand side and to values on the z-axis, i.e. represents glucose (a) and lactate (b) concentrations.
