NHERF1 time-dependently interacts with FATP5 in the hepatic membrane fraction. (a) Western blot analysis of FATP5 and EZRIN in immunoprecipitates (IPs) from hepatic membrane fractions using anti-NHERF1 antibodies. (b) FATP5 protein levels on the plasma membrane prepared from NHERF1-expressing Hepa1–6 cells. Cells were transfected with plasmid vectors encoding Slc9a3r1 (Slc9a3r1 vec.). Control groups were transfected with empty vectors (pcDNA3.1) instead of expression plasmids. Presence (+) or absence (-) of expression plasmids (0.5 μg each) is denoted. Coomassie Brilliant Blue (CBB) indicates the equal loading of proteins from the membrane fraction. Each value represents the mean with s.e.m. (n = 6). **P < 0.01, significant difference from control group (t10 = 6.130 for FATP5; t10 = 3.919 for NHERF1; unpaired t test, two-sided). (c) The transporting activity of FATP5 in NHERF1-expressing Hepa1–6 cells. Each value represents the mean with s.e.m. (n = 6). **P < 0.01; *P < 0.05, significant difference from control cells at the corresponding times (F5,50 = 48.556; P < 0.001; ANOVA with Tukey-Kramer post hoc test). Full-size images of immunoblotting are presented in Supplementary Figs 15 and 16.
