The effect of ODN 2216 stimulation on activation of TLR9 signalling in CD4+ Teff cells. CD4+ Teff cells were stimulated using ODN 2216 for 14 hr and its effect on expression of genes involved in TLR9 signalling was assessed by real time RT-PCR. We observed a significant decrease in mRNA levels of (a) TLR9 (p = 0.01), (b) MyD88 (p = 0.02), (c) IRAK1 (p = 0.05), (d) TRAF3 (p = 0.04), (e) IRF7 (p = 0.03) and (f) CHUK (p = 0.03) in subjects with IOTB as compared to non-uveitis controls. However, few genes of TLR9 signalling pathway including (g) IRAK4 (p = 0.11) and (h) TRAF6 (p = 1.0) showed no significant differences in expression levels.
