PI3K and WNT inhibitors induce a mitotic delay. (A,B) HeLa M cells were synchronized by single thymidine block and then exposed to 100 nM Taxol and either 2.8 μM LY294002 (LY) or 400 nM WNTc59. Cells were harvested after 13 hours in HCT116 or 16 hours in HeLa cells, and chromosome spreads used to analyze mitotic index. ~300 cells per condition was counted in a blinded manner, and the experiments performed in triplicate and repeated three times. (C) HeLa M cells or HCT116 cells (D) were synchronized by single thymidine block and then exposed to 100 nM Taxol and either 2.8 μM LY294002 (LY) or 400 nM WNTc59, or both and traced using time lapse analysis. Mitotic length was measured using mitotic entry and exit times, with 30 cells being counted for each treatment.
