Scheme of plant material collection to obtain APEX, iFIB, FIB samples for subsequent RNA-Seq analysis. Bundles of the intrusively growing fibers (sample iFIB) were isolated by laser microdissection from the longitudinal cryosections of 3rd cm from the stem apex. Sample APEX was collected as 2 mm of the uppermost stem together with leaf primordia. The described earlier12 sample FIB (fibers isolated during tertiary cell wall deposition) was collected from the stem portion below the snap point (SP).
