The proposed positive feedback mechanism for Th17 cell-secreted IL-17A and NLRP3 inflammasome activity in a circuit. In this proposed mechanism, specific natural compounds, such as luteolin, and wedelolactone, exhibited anti-colitis activity and suppressed IL-17A expression in Th17 cells and IL-17R-activated signaling at the post-transcriptional level, which further activated the specific TLR pathway and NLRP3 inflammasome. We also included the possibility that these phytochemicals can suppress the expression of specific inflammasome components, such as NLRP3 and NLRP1, via yet unknown mechanisms.
