14-3-3, PI4KB and Rab11 form complex of 2:1:1 stoichiometry. (A) Schematic representation of the domain composition of the proteins used in the study. PI4KB is composed of the N-terminal region, helical domain, and kinase domain which can be divided into N- and C-terminal lobes. Phosphorylation site at S294 is indicated. ACBD3 contains the acyl-CoA binding domain (ACBD), charged amino acids region (CAR), glutamine rich region (Q), and Golgi dynamics domain (GOLD). Rab11 is composed of the N-terminal GTP-binding domain and disordered C-terminus which can be geranylgeranylated at C212 and C213 as indicated. (B,C) c (S) distribution profiles revealing that P-PI4KB:Rab11 complex is preferentially formed with 1:1 stoichiometry (B) and 14-3-3:P-PI4KB:Rab11complex with 2:1:1 stoichiometry (C). Sedimentation coefficients of all complexes analyzed by AUC are listed in SI Table 1.
