Effect of TLR2 and TLR4 verified by siRNA and receptor knockout model. (A–C) rVSMC were transfected with siRNA for TLR2 (TLR2siRNA), TLR4 (TLR4siRNA) or both (TLR2/4siRNA) as well as negative control siRNA (NC). Mock transfected cells were used as control in all experiments. Afterwards, cells were stimulated for 24 h with rhSAA (1 µg/ml), LPS (1 µg/ml) and HKSA (107 cells/ml) with subsequent determination of MCP-1 mRNA expression.
