The expression of GluN2B or phosphorylated GluN2B at Y1472 in the ARC area following the CFA injection into the hind paw of rats. The gel shown in (a) was loaded with lysates prepared from the hypothalamic ARC area of animals without any treatment (Naïve) or at days 1, 3, 7 and 14 after the CFA injection into the hind paw. The group of blots were cropped from a same filter, and stripped and successively probed with an antibody against GluN2B-pY1472 (2B-pY1472, top blot), GluN2B (middle blot) or GAPDH (bottom blot). The ratio of the band intensity of GluN2B-pY1472 or GluN2B versus that of GAPDH was calculated. Bar graphs in (b) show normalized ratios to those in naïve animals ( = 1). The bar graph in (c) shows the ratios of the band intensities of GluN2B-pY1472 versus GluN2B. *, **p < 0.05, p < 0.01, Dunnett’s post hoc test in one-way ANOVA in comparison with those of naïve rats. Values in brackets indicate the number of experimental repeats.
