Cytokine and chemokine secretion profiles of resting human Vδ1+ T cells obtained from PBMCs stimulated with hesperidin and linarin. (A) The percentage of Vδ1+ T cells among PBMCs were determined as 0.47%, and the Vδ1+ T cells were sorted by FACSAriaII and were expanded with 2 μg/mL PHA and 100 U/mL rIL-2. The open histogram indicates staining with anti-Vδ1 mAb, and the shaded histogram shows the staining with the isotype control. The stained cells were analyzed by FACSCantoII using FlowJo software. (B) The cytokine profile of culture supernatant from the resting Vδ1+ T cells stimulated with/without 25 ng/mL PMA and 1 μg/mL ionomycin (P.C.), 100 μg/mL hesperidin or linarin, or 0.01% DMSO for 3 days. The amounts of these cytokines were measured by BD™ CBA kit and FACSCantoII. The data are expressed as the mean + SEM (n = 4). *P < 0.05 ANOVA with Dunnett’s test. (C) IL-5 and IL-13 (upper) and MIP-1α, MIP-1β and RANTES (lower) amounts of culture supernatant from the resting Vδ1+ T cells stimulated with/without hesperidin, linarin, DMSO for 3 days. The amounts of these cytokines and chemokines were measured by using each specific ELISA kit.
