From: Establishment and Characterization of a New Cell Line Permissive for Hepatitis C Virus Infection
Replication of several HCV genotypes. H77S.3/GLuc2A (genotype 1a), HJ3-5/GLuc2A (genotype 1a/2a chimaera), N2/GLuc2A (genotype 1b), and JFH1/GLuc2A (genotype 2a) RNA were transfected into KH and Huh-7.5 cells. A replication-deficient mutant, H77S/GLuc2A/AAG, was also transfected. Medium was replaced at 6 h and then at 24 h intervals until 96 h. Secreted GLuc activity was determined and normalised to that at 6 h. GT denotes genotype. Differences in the means of normalised GLuc activity between KH and Huh-7.5 cells at each time point were analyzed by Student’s t test.
