Effect of HCV infection on ISG mRNA levels. HCVcc derived from HJ3-5 was used to infect Huh-7.5 and KH cells at a multiplicity of infection of 1. Total cellular RNA was extracted at regular intervals after infection, and HCV RNA and mRNA levels of ISGs, such as MX1, SOCS3, IFNβ, and OAS2, together with β-actin for normalisation were quantified using qRT-PCR. Differences in the means of mRNA levels between KH and Huh-7.5 cells at each time point were analysed with Student’s t test.
