Detection of DNA double-strand breaks via γH2AX staining in LN18 cells after treatment with 213Bi-anti-EGFR-MAb. Cells were incubated with 213Bi-anti-EGFR-MAb for 3 h at 4 °C to inhibit DNA-repair or mock-treated with PBS (controls). Subsequently cell culture medium was exchanged and cells were incubated for 1 h at 37 °C to initiate DNA-repair. Shortly thereafter, cells were fixed, stained for γH2AX (B,E) and counterstained with DAPI for visualization of the nuclei (A,D). Intranuclear foci indicate DNA double-strand breaks induced by the alpha-emitter 213Bi (C,F overlay, representative images).
