Differential nitric oxide (NO) production by peripheral blood mononuclear cells (PBMCs) isolated from specific pathogen-free chickens and stimulated with LPS. Birds were immunized at 4 weeks of age by ocular instillation of live intermediate plus IBDV vaccine and intramuscular injection of Pam3CSK4 and/or poly I:C. PBMCs (n = 6/group) were isolated on 21 dpi and stimulated using LPS (10 μg/ml) keeping medium control. The production of NO was analyzed using Griess reagent as a nitrite at 24 h post-stimulation, and the difference between stimulated and unstimulated cells in the same bird was calculated. Treatment effects were analyzed by one-was analysis of variance. The probability of an alpha error was set at 0.05.
