Expression characteristics of H4_GE cells – protein amounts depending on tebufenozide concentration. H4 cell lines were incubated for 4 days with DMSO or different tebufenozide concentrations ranging from 10 nM to 10 µM. For all samples, expression of housekeeper β-tubulin was used as reference. (A) Control Western blots of H4 cells were treated like the Western blots of the corresponding samples and showed no expression of αSyn, Venus, SV, V1S, or SV2. (B) In all transgene expressing cell lines signal intensity increased with increasing tebufenozide concentration up to 1 µM or 10 µM for inducible transgenes. Full-length Western blots and additional exposures are shown in Supplementary Figs 5, 6, respectively. (C) Quantification of Western blot signal intensity was performed using ImageJ. Signal intensity of transgenes was normalized to maximum. Data points were derived from the blots shown in A and B.
