LPS-induced transcript expression of IL-10 and IFN-β is controlled by the interplay of MK2 and MK3. Bone marrow-derived macrophages (BMDM) were prepared from wild-type (wt), MK2-deficient (MK2−/−) and MK2/3-double-deficient (MK2/3−/−) mice and treated with 100 ng/ml LPS for the indicated durations. Total mRNA was extracted and analyzed using rtPCR. Data are presented as the means ± SEMs based on at least 5 replicates. Statistics were calculated by 2-way ANOVA with the Bonferroni test. A p-value of less than 0.05 was considered significant. The symbols indicate significance: *) wt vs. MK2−/−, +) wt vs. MK2/3−/− and x) MK2−/− vs. MK2/3−/−.
