(a) The 2D algorithm reproduces the read-out of γ-H2AX foci in ICC images taken with 2D microscopy through: selection of a slice of thickness Δz (centered in the middle of the nucleus) (1); planar projection of DSBs (DSB clusters) within the slice (2); merging in a single focus of DSBs (DSB clusters) falling within a clustering radius r (3). (b) The 3D algorithm reconstructs the yield of foci in the whole nucleus by merging in a single focus DSBs (DSB clusters) falling within a clustering radius R.
