Cell sheet experimental protocol. Human umbilical cord stem cells (hUC-MSCs) were seeded on temperature responsive cell culture dishes (TRCD) and cultured to confluence in a 37 °C cell culture incubator. Cultured cells were detached from TRCDs as intact cell sheets within 20 min at room temperature (RT), preserving their functional structures such as extracellular matrixes (ECMs) and cell-cell-junctions and cytokine secretion ability without using proteolytic enzyme treatments.
