Measurement of the half maximal inhibitory concentration (IC50) of CX-4945 for hsCK2α and cnCka1 through in vitro kinase assay. In vitro kinase assay of (a) human CK2α and (b) C. neoformans Cka1. The recombinant proteins were incubated with CX-4945, respectively, and consumed ATP by each kinase activity. The produced ADP was converted to ATP to be used by luciferase, and the generated luminescence was detected by a luminometer. The counts per second (CPS) is indicative of the kinase activities of hsCK2α and cnCka1. Three-independent experiments with three-technical replicates were performed and represented in each logarithm graph. The error bar indicates ± S.E.M., and the R-square values are as follows: hsCK2α (0.9654); cnCka1 (0.9962).
