Relative gene expression changes after radiation in human and mouse cells 24 hours after irradiation (opposite response patterns). Genes were selected from microarray analyses for showing opposite responses (at least in one condition) to radiation in human and mouse blood. Values are mean fold change of n > 5 biological replicates, for four experimental groups Human (Human blood irradiated ex vivo, pink line and symbols), HuNSG (Humanized NSG mice whole body irradiation, human genes, purple line and symbols), C57BL/6 (mice after whole body irradiation, light blue line and symbols) and NSG (age-matched NSG non-engrafted mice after whole body irradiation, dark blue line and symbols). The grey line indicates the calibrator sample, which is the matched sham-irradiated sample for each experimental group. For the PTPRO gene, the relative expression of human genes is shown as it agreed with the response as measured by microarrays, up-regulated after radiation, however, the levels of this mRNA were undetectable by qPCR in the mouse samples, C57BL/6 and NSG, therefore are not shown. Most of the NSG transcripts (dark blue) for the selected genes followed the expected C57BL/6 response (light blue) except for the KCNN4 gene which was up-regulated, as in the human response.
