Crystal structures of active RSK2 and Akt2/PKBβ show the interaction between the phosphate group on the AL and three positively charged amino acids. (A) The crystal structure of N-terminal wild type RSK2 kinase in complex with 2-amino-7-substituted benzoxazole (PDB ID 4NW5) shows the interactions between pSer227 and other three positively charged amino acids: Arg114, Arg192 and Lys216. (B) In the same structure of RSK2, pS227 was substituted with an Asp. (C) Structure of the active AL in Akt2/PKBβ (PDB ID 1O6K), where Thr309 interacts with Hys196, Arg274 and Lys298.
