Identification of the key enzymes involved in histone methylation and DNA methylation. (a) HMT and DNMT bands were detected by Western blot, and the enzymes assessed were G9A, Suv39h1, DNMT-1, and DNMT-3a. Images were cropped for clarity, and full-length blots/gels are presented in Supplementary Fig. 2. (b,c) Quantitative analysis of these enzymes. In the process of inducing differentiation, G9A expression decreased gradually, DNMT-3a expression increased, and the other enzymes were not obviously changed. (d,e) The binding levels of these enzymes in the GATA4/Nkx2.5 promoter were detected by ChIP-qPCR; a normal mouse IgG antibody used as a negative control. The input% showed that the main HMT and DNMT binding in GATA4/Nkx2.5 promoters was G9A and DNMT-1, respectively. *p < 0.05 compared with the blank group.
