BaP1 induces production of IL-1β, which contributes to PGE2 production. (A) Time-course of BaP1-induced IL-1β release. FLS monolayers were incubated with BaP1 (12.5 µg/mL) or RPMI (control) for 30 min, 1, 3, 6, 12 and 24 h. (B) Isolated FLSs were pretreated with Anakinra (50 μM) or vehicle and stimulated by BaP1 (12.5 mg/mL) or RPMI (control) or IL-1β for 12 hours. Supernatants were collected, and IL-1β and PGE2 were quantified by ELISA. Results are expressed as mean ± SEM of 4 samples. *p < 0.05 relative to control. #p < 0.05 relative to BaP1 (12 h); &p < 0.05 relative to BaP1; +p < 0.05 relative to IL-1β (two-way ANOVA, Bonferroni post test in A and one-way ANOVA, Bonferroni post test in B).
