Extract of roasted coffee beans suppresses LPS-induced NO production and expression of iNOS. RAW264.7 cells were pretreated with various concentrations of coffee extract (1.25, 2.5, and 5% (v/v)) for 1 h prior to the stimulation with LPS (1 μg/mL). (A) The nitrate concentrations in culture supernatants were measured 24 h after the LPS stimulation using Griess reagent. **p < 0.01, ***p < 0.001 significantly different from control cells treated with LPS. (B) The cell viability was analyzed 24 h after the LPS stimulation using trypan blue exclusion tests. (C) iNOS mRNA expression was evaluated 12 h after the LPS stimulation by RT-PCR. GAPDH mRNA expression was used as an internal control. **p < 0.01, ***p < 0.001 significantly different from control cells treated with LPS.
