Susceptibility of EBOV replication to tenatoprazole. HeLa cells were pre-treated for two hours with media containing either DMSO (0) or ten concentrations of tenatoprazole up to 100 μM in DMSO. EBOV (Zaire) was added at MOI = 0.5. After 24 hr, infection was stopped by fixing the cells and virus spread was determined by immuno-staining for viral glycoprotein (GP). Panel A, Triplicate samples showing GP-staining signal after 24 hr. Left, cells treated with DMSO alone; Right, cells treated with 100 μM tenatoprazole in DMSO. Scale bars equal 50 microns. Panel B, Inhibition of virus infection quantified based on reduction in virus spread determined at each concentration (N = 8 for each concentration). Cell viability was determined from the number of nuclei per well in treated samples compared to infected but untreated controls. Virus spread was calculated relative to cells with no drug. Error bars equal 1 SD.
