Ex vivo Inhibition of serine racemase. (A) Cultured hippocampal neurons were treated with vehicle or madecassoside and stained with D-serine antibody to visualize D-serine production. Note the D-serine puncta in the neuronal soma and the processes in the vehicle-treated neurons (bottom). However, the total number of puncta significantly disappeared upon madecassoside treatment (top) indicating an inhibition of SR. Two independent cultures were used. (B) Quantification showing significant reduction of D-serine fluorescence in madecassoside treated cell compared to control cells.
