IRS4PVH neuron activity is dispensable for melanocortin agonist-mediated satiety. In situ hybridization for Irs4 (A, blue) and Mc4r (A, red) mRNA in the PVH of adult wildtype mice. Melanocortin agonist-induced satiety was tested in mice with IRS4PVH neuronal silencing (B,C). Two-hour (B) and four-hour (C) feeding was significantly decreased following i.p. injection of the MC4R agonist MT-II (150 ug/mouse) in all cohorts of mice, despite the silencing of IRS4PVH neurons in the PVHTetTox group. Average values ± SEM are shown, **p < 0.01, ***p < 0.001, ****p < 0.0001; PVHTetTox n = 6, PVHFlex n = 17, WTTetTox n = 13.
