(A) Differentiated cells were incubated with 10 nM bafilomycin A1 (BAF) for 6 h or with equivalent amount of DMSO as vehicle (UNTR), and Western blotting was performed for LC3A, LC3B and p62. Total (B) LC3A (LC3A-I + LC3A-II) and (C) LC3B (LC3B-I + LC3B-II) levels were normalized to actin. (D) LC3A-II, (E) LC3B-II and (F) p62 levels for each group were calculated by densitometric analysis and normalized to actin. Full-length blots are presented in Supplementary Fig. S4. (G) Total LC3-II flux and (H) p62 flux calculated as the ratios LC3-II (BAF)/LC3-II (UNTR) and p62 (BAF)/p62 (UNTR), respectively.
