Effects of chemotherapeutics on apoptosis in HL-60 cells. Cells were treated with 11.3 µM cisplatin, 20.7 µM PyTT, 20.7 µM PdPyTT, or the vehicle (DMSO, control) for 24 h. (A) Hoechst 33342-stained cells were visualized with epifluorescence microscopy and representative images of each experimental condition are shown. The fraction of apoptotic nuclei was evaluated as indicated in the Materials and Methods section. Yellow arrows indicate apoptotic nuclei (i.e., fragmented or condensed). Both fragmented and condensed nuclei are also shown in greater detail in the magnifications. (B) Histograms show percentages of cells with apoptotic nucleus.
