(a) Type I collagen is cast into cushions in a 96-well microtiter plate; (b) Single-donor human primary endothelial cells are grown over the cushions until achieving confluency (representative image); (c) CD33-purified human monocytes (representative histograms for CD14 expression) are then added to confluent tissue constructs (TCs) to enable autonomous extravasation for 1.5 h at 37 °C/5%CO2; (d) non-migrated cells are then removed by gentle aspirations; (e) TCs are then examined by microscopy or digested to extract extravasated monocytes for further analysis.
