Rapid, millimeter-scale, time-resolved SPC, MPM ex vivo imaging of fresh scalp tissue. Tile-mosaic images (4.5 × 4.5 mm2, 25 MPx) acquired and restored with CARE in 2.5 min at select depths of 30 μm (a), 50 μm (b) and 80 μm (c) corresponding to the epidermis, DEJ and papillary dermis, respectively. Mosaic images are color-coded by short-lifetime TPEF (red), long-lifetime TPEF (green) and SHG (blue). (d,e,f) Digital zoom to the 1.8 × 1.8 mm2 white outlined areas in (a), (b) and (c), respectively, showing tissue heterogeneity on mm scale. (g,h,i) Digital zoom to the 450 × 450 μm2 white outlined areas in (d), (e) and (f), respectively, showing non-pigmented keratinocytes and elastin fibers predominantly visualized in the green channel, pigmented (melanin-rich) keratinocytes mainly detected in the red channel and collagen fibers visualized through the SHG signal in the blue channel.
