Impact of 2E3 binding reactivity on prozone effect observed in the SAB assay. The MFI difference (∆MFI) between 2E3 tested at 2 μg/mL and 2E3 tested at 10 μg/mL was calculated for each allele and plotted according to the 2E3 reactivity categories as classified in Table 1 and different 2E3 subclasses using (A, B) Pan-IgG detection reagent, and (C, D) corresponding subclass-specific detection reagents. Data represented as mean with 95% CI. Statistical significance between reactive (N = 15), cross-reactive (N = 18) and non-reactive categories of the same subclass (N = 64) tested at the same center was calculated by one-way ANOVA Kruskal–Wallis tests followed by Dunn’s multiple comparisons test.
