Hif1α activation impairs cDC function. (a) Antigen presentation capacity and activation of DFO-treated cDCs was evaluated by MLR. When indicated, cells were also treated with the fatty-acid synthesis inhibitor TOFA. T cell proliferation was assessed by CFSE dilution by flow cytometry. Histograms are representative of 3 independent experiments. Bar graph indicates proliferation index analysed by flow cytometry. (b) Cells were treated overnight with DFO and then stimulated with LPS for 6 h or overnight. Intracellular IL-12 production was evaluated by flow cytometry 6 h after stimulation. Dot plots are representative of 3 independent experiments. (c) Cytokines were measured in the supernatant WT cDCs after overnight stimulation by ELISA (n = 3). Statistical analysis was performed with one-way ANOVA, **p < 0.01 and ***p < 0.0005.
