Imaging of optically cleared Mtb infected tissue. (A) Photographs of Mtb infected tissue before and after the scaleCUBIC clearing process. For scale, the NIBSC logo with a diameter of 13 mm is presented behind the cleared lung lobe which was approximately 2 mm thick. Approaches to image optically cleared tissue are presented with imaging by (B) a 1.25x /0.04NA lens and (C) a 20x /0.75NA glycerol immersion lens. In Mesoscopy (D) cellular detail can be seen and (E) over a large field of view in 3D without the need to stitch the images. Samples infected with Mtb (not specifically stained) and stained with propidium iodide (nucleic acid) for CLSM and Mesoscopy. All images are maximum intensity projections of Z stacked images.
