The effect of the knockdown of YBX2. (a) The level of YBX2 protein expression was investigated by Western blotting. Full length blots are shown in Supplementary Fig. S5b online. (b) The level of YBX2 gene expression using two siRNAs (No.1 or No.3) was investigated by real-time PCR. GAPDH level was used as internal standards. Relative ratios are represented as the means ± SD from three independent experiments (*p < 0.05). (c) Downregulation of YBX2 protein using two siRNAs led to significant decreases in the population of SP cells compared with that using control siRNA. (d) The percentage of SP cells was decreased in IK-YBX2 C1 siRNAs cells compared with IK YBX2 C1 control siRNA cell from three independent experiments (*p < 0.05). (e) The level of ALDH1 expression was decreased in IK-YBX2 C1 siRNAs No.1 and No.3 compared with IK-YBX2 C1 control siRNA cell. HPRT level was used as internal standards (*p < 0.05).
