Effect of the D797N/K799N mutation on the ER-stress responsiveness of ΔIΔIIIΔV/Y225H Ire1. After being grown at 30 °C in SD medium, KMY1015 cells (ire1Δ) carrying the indicated mutant versions of the IRE1 plasmid pRS313-IRE1 were ER-stressed and checked for HAC1-mRNA splicing efficiency using the RT-PCR technique. (a and b) Cells were cultured in the presence of 2 µg/mL tunicamycin for the indicated durations. (c) Cells were cultured in the presence of 2 µg/mL tunicamycin for 2 h or remained unstressed. (d) Cells were cultured in the presence of 3 mM DTT for the indicated durations. (e) Cells remained unstressed or were cultured in the presence of 1 or 3 mM DTT for 60 min or 10 mM DTT for 30 min.
