P. vivax isolation, short-term ex vivo maturation, enrichment and total RNA extraction. Blood samples were immediately processed. White blood cells were depleted by CF11 filtration25. The early blood staged parasites were short-term cultured ex vivo to allow maturation followed by Percoll gradient enrichment23. Total number of Pv-iRTs was accessed before preservation in RNAlater or TRI Reagent at − 80 °C. RNA extraction was executed using the RNeasy Micro kit.
