Inhibition of cPLA2 with PACOCF3 reduces the pro-inflammatory effects of myelin on M1 macrophages. BMDMs were grown and stimulated as in Fig. 2, with or without the addition of cPLA2 inhibitor, PACOCF3 (50 µM). (A) cPLA2 inhibition reduced myelin mediated ROS increases in M1 (IFN-γ and LPS) macrophages. (B) cPLA2 inhibition reduced myelin mediated nitric oxide production. (C) cPLA2 inhibition reduced the neurotoxic potential of M1 macrophages treated with myelin as determined by an MTT assay measurement of N2a cell viability. (D) cPLA2 inhibition did not significantly alter arginase activity. Representative of 3 biological replications of both BMDMs and myelin source. *p < 0.05, **p < 0.01, ***p < 0.001 mean ± SEM.
