Isothermal titration calorimetry analysis of Ni binding to Aβ40. Top panel shows the raw data of heat release per injection, for 20 consecutive injections (2.38 µL) of NiSO4 (1 mM) into a 500-µL cell containing Aβ40 (20 µM). Bottom panel shows binding isotherms, obtained by integrating the areas of each injection peak. Data acquired with a NanoITC were analyzed using NanoAnalyze 1.2 software(TA Instruments). Shown in the inset are the best-fit values for the dissociation constant (Kd), stoichiometry (n), enthalpic change (ΔH), and entropic change (ΔS).
