Ex vivo production of lung epithelial microvesicles (MVs). Human lung specimens (pieces from the same lung) were stimulated with PLY (5 μg/ml) or PBS (Ctr). MVs were isolated from the conditioned media 8 h later and analyzed by FACS. (A) Representative overlay dot blot of MVs from PLY-treated human lungs. Blue dots represent MVs double stained with isotype and annexin V in the presence of EDTA (negative staining). Red dots represent MVs double stained with annexin V-APC and EPCAM-Alexa488. Epithelial MVs (double positive events) are gated by the ellipse gate (green). (B) Graph depicts the quantification of annexin V/EPCAM double positive MVs.
