(A) A schematic of KASH5 TMD indicating the sequential replacement of a leucine with a glutamine in the 7-leucine stretch. Computed according to the GES scale, all the variants have the same hydrophobicity of the TMD. (B) MCF-7 cells transfected with a N-terminal GFP-tagged version of KASH5 variants and treated with MitoTracker Red CMXROS to stain the mitochondria. All substitutions within the 7-leucine stretch lead to the mislocalization at the mitochondria. Scale bar 10 μm. The experiment was repeated two times.
