Hydrophobicity of the transmembrane domain alters KASH5 protein localization. Using the GES scale to calculate the hydrophobicity, the L535V and L535N substitutions mimic the hydrophobicity of the TMD of the wild-type and L535Q KASH5, respectively. (B) MCF-7 cells transfected with a GFP-tagged version of wild-type or variant KASH5 and treated with MitoTracker Red CMXROS to stain the mitochondria. Both wild-type and L535V KASH5 localize at the nuclear envelope while both L535Q and L535N localize at the mitochondria. This experiment was repeated four times.
