Hydrophobicity of the transmembrane domain alters KASH5 protein localization. (A) Schematic representation of KASH5 TMD indicating the different amino acid substitution at the position 535 (V, valine; Q, glutamine; N, asparagine). Using the GES scale to calculate the hydrophobicity, the L535V and L535N substitutions mimic the hydrophobicity of the TMD of the wild-type and L535Q KASH5, respectively. (B) MCF-7 cells transfected with a GFP-tagged version of wild-type or variant KASH5 and treated with MitoTracker Red CMXROS to stain the mitochondria. Both wild-type and L535V KASH5 localize at the nuclear envelope while both L535Q and L535N localize at the mitochondria. This experiment was repeated four times.
